SAMe Participates in Over Forty Essential Biochemical Reactions


SAMe (pronounced Sammy) is an acronym for S-adenosyl-L-methionine, which is normally produced in the liver from the amino acid methionine. In the body, SAMe functions as a methylation agent, transferring methyl groups between molecules. Although other substances donate methyl groups, SAMe is the most active of all methyl donors. It participates in 35-40 essential biochemical processes including detoxification reactions and manufacture of brain chemicals, antioxidants and joint tissue structures.(1,2)

These actions make SAMe an effective nutritional support supplement for a variety of conditions including:

  • Depression

  • Fibromyalgia

  • Osteoarthritis

  • Liver Disorders

  • Male Infertility


  • Folic acid and vitamin B-12 are necessary for the synthesis of SAMe, and deficiencies of these vitamins result in low concentrations of SAMe in the central nervous system (CNS).(2) Low blood or CNS levels have been detected in people with cirrhosis of the liver,(4) coronary heart disease,(5) Alzheimer’s disease and depression.(3)

    Safe, Fast, Effective Mood Enhancement Supplement

    Supplemental SAMe has been shown to possess antidepressant activity with a rapid mode of onset and minimal side effects.(4,5) According to Dr. Richard Brown, an associate professor of clinical psychiatry at Columbia University, "SAMe is faster, better and has hardly any side effects" compared to prescription antidepressants.(5)

    Researchers began investigating SAMe as an antidepressant in the 1970’s. Controlled studies suggest that 70% of depressed patients respond to SAMe -- roughly the same proportion that benefits from any established drug treatment -- and SAMe tends to work more quickly. Many patients report noticeable effects within one week.(6,9) SAMe has also been found effective in speeding the onset of action of antidepressant drugs. A double-blind clinical trial involving 40 outpatients with moderate to severe depression combined SAMe with imipramine (IMI). Depressive symptoms decreased earlier in the SAMe-IMI group than those in the placebo-IMI group.(8)

    Another double-blind trial compared SAMe to IMI. By the end of the second week, 66% of the SAMe patients showed clinically significant improvement in depressive symptoms, compared to 22% of the IMI patients.(9)

    Some really interesting results were seen in a double-blind trial at University of California Irvine Medical Center when SAMe was compared to the drug desipramine. At the end of the four week trial, 62% of the patients treated with SAMe and 50% of the patients treated with desipramine had significantly improved. However, all patients who showed a 50% decrease in their Hamilton Depression Scale score, also had a significant increase in plasma SAMe, regardless of whether they were treated with SAMe or the drug.(7,10)

    Studies show that SAMe can increase levels of the important neurotransmitters serotonin, norepinephrine and dopamine. It also influences the metabolism of phospholipids such as phosphahtidyl choline and phosphatidyl serine. SAMe may enhance the impact of serotonin and dopamine, either by regulating their breakdown or by speeding production of their receptor molecules. It may also make existing receptors more responsive. By methylating phospholipids, it keeps membranes fluid and receptors mobile.(7)

    Relieve Pain and Promote Joint Health

    When some of the first patients given SAMe for depression also reported improvements in their joint pain, researchers saw another potential use. Over 22,000 arthritis sufferers have now been through studies comparing SAMe to placebos and to non-steroidal anti-inflammatory drugs (NSAIDs) such as aspirin and ibuprofen. After four weeks of treatment, relief from pain, stiffness and swelling using SAMe is equal to that of NSAIDs. Other studies suggest that the compound may restore damaged cartilage. When SAMe is broken down it generates other valuable molecules, including sulfate groups that help maintain cartilage.(6,7)

    Liver Disorders, Migraines, Fibromyalgia and Infertility

    Studies suggest that SAMe can help normalize liver function in patients with cirrhosis, hepatitis and blockage of the bile ducts. It has also been found to prevent or reverse liver damage caused by certain drugs. Patients with cirrhosis may have an acquired metabolic block in the hepatic conversion of methionine to SAMe. Cirrhosis can be caused by excess alcohol consumption. Since alcohol depresses an enzyme needed to convert methionine into SAMe, supplementation can help regenerate normal liver function.(11)

    Intravenous SAMe given to fibromyalgia patients reduced pain and depression in two double-blind studies.(12,13) In another double-blind study, oral supplements significantly reduced pain, fatigue and stiffness and improved mood.(14)

    Preliminary research also suggests that oral SAMe may increase sperm activity in infertile men(15) and may be useful in treating migraines.(5,16)

    Vitamins B-6, B-12 and Folic Acid

    The synthesis of SAMe is intimately linked with folate and vitamin B-12 metabolism. Deficiencies of both of these vitamins have been found to reduce SAMe concentrations in the central nervous system. Vitamin B-6 is an important coenzyme in the synthesis of all neurotransmitters and levels are typically quite low in depressed patients, especially those using oral contraceptives.(17)

    These B vitamins are even more important after SAMe has completed its methylation process. When a SAMe molecule gives up its methyl group, homocysteine is formed. Excess homocysteine is a major risk factor for heart attack and stroke and has been implicated in depression. When B-6, B-12 and folic acid are available, homocysteine is easily converted back into methionine or into glutathione, a valuable antioxidant.(7) SAMe X-Tra includes these vitamins to ensure that homocysteine is converted into beneficial substances rather than being allowed to build up.

    Recommended Use

    Because SAMe is absorbed mainly through the intestine, it’s best taken in enteric coated tablets that pass through the stomach intact. In research studies involving a variety of conditions SAMe has been used in the following amounts:

    Depression - 200 to 1600 mg. daily, depending on the degree of depression.

    Osteoarthritis - 400 to 1200 mg. daily.

    Fibromyalgia - 800 mg. daily.

    Liver Disorders - 1200 mg. daily.

    Migraine - 800 mg. daily.

    This supplement is not recommended for use by manic-depressive (bipolar) patients.
